Day 1: Adelaide

Arrive in leafy Adelaide and spend some time shopping and sight seeing - the many parks are pleasant throughout the day, and in the evening, there are dozens of places to have a few drinks under the pretension of sampling the local wines.
Hotel: Majestic Roof Garden Adelaide Hotel

Day 2: Adelaide - Goolwa

Distance: 90 Miles
Take a leisurely breakfast and then hop in your car to drive out to the McLaren Vale wine-producing region. Take the chance to sample a few glasses of the local specialities before you head on to Goolwa for an eco cruise into the Coorong National Park on board the "Spirit of Coorong". When the sun sets, spend some time soaking up the silence of Goolwa over a glass of wine. I recommend the excellent Barossa.
Hotel: Goolwa Self Catering Cottage

Day 3: Goolwa - Kangaroo Island

Distance: 60 Miles
Just off the coast of South Australia, Kangaroo Island has many attractions and is well worth spending a couple of days exploring. Set off early and get the ferry from the mainland. Check out Stokes Bay, Seal Bay, and the Emu Ridge Eucalyptus Distillery before going back to your hotel for the evening.
Hotel: Kangaroo Island Hotel

Day 4: Kangaroo Island - Adelaide

Distance: 25 miles
Get up nice and early to see some of the many natural sights on the island - highlights include the so called "Remarkable Rocks", which are, well, remarkable, Kelly Hill Caves, and the admiral's Arch. When you've had your fill of geological marvels, head for the ferry terminal and go back to the city to spend the evening searching for the perfect cocktail bar or pub.
Hotel: Majestic Roof Garden Adelaide Hotel

Day 5: Adelaide

Spend the day sight seeing in Adelaide, before returning your hire car and boarding your plane onwards.
